A dilution series of Escherichia coli has been prepared, seeded into the PCA and incubated aerobically at 25° C for three days. E. coli is a facultative anaerobic. Colonies on the surface is relatively large, off-white, while the colonies, which are inside the agar is lenticular. Colonies which lies between the Petri dish and the agar is transparent and gray.

Colonies located on the surface are relatively large, off-white and round. Colonies which lies in the agar are whitish and lenticular. Colonies which lies between the agar and the petri dish are circular and greyish, transparent.
